Professor Luo Xianghang's Team Publishes an Original Article about Exercise-Stimulated Bone Formation and Immune Response

On June 15, the research team of Professor Luo Xianghang, Director of Department of Endocrinology, Endocrinology Research Center, Xiangya Hospital of Central South University, online published an article about exercise-stimulated bone formation and immune response entitled “A Mechanosensitive Lipolytic Factor in the Bone Marrow Promotes Osteogenesis and Lymphopoiesis” in Cell Metabolism (IF=27.287). Physician Peng Hui from Department of Endocrinology, Xiangya Hospital of Central South University is the first author, and Professor Luo Xianghang is the corresponding author.



Exercise can prevent osteoporosis and improve immune function, but the mechanism remains unclear. Researchers found that there is the crucial role of lipolysis in exercise-stimulated osteogenesis and lymphopoiesis; exercise promotes reticulocalbin-2(RCN2) secretion from the bone marrow macrophages .and macrophage-specific RCN2 ablation impaired exercise-stimulated osteogenesis, lymphogenesis and antiviral ability; RCN2, as a new lipolytic factor,  binds to a functional receptor complex, which is composed of neuronilin-2 and integrin beta-1, to activate a cAMP-PKA signaling pathway that mobilizes bone marrow fat via lipolysis to fuel the differentiation and function of mesenchymal and hematopoietic stem cells. The team further synthesized the recombinant RCN2, which was shown to significantly alleviate bone loss and improve lymphopoiesis in mice. Currently, recombinant RCN2 has applied for a national invention patent.


Professor Luo Xianghang's team has long been dedicated to clinical and basic research on endocrine metabolic diseases. This study has resolved the molecular mechanism of exercise-stimulated bone formation and immune response, revealed a new theory of bone marrow adipose tissue as an energy storage reservoir to fuel bone formation and immunity response, and brings new hope for the prevention and treatment of osteoporosis and improvement of immunity.
